Colchester's Essex neighbours Chelmsford produced an FA Cup first round giant killing act to end The U's involvement for another season.
Midway through the first half a fine strike from Donovan Simmonds (23) gave Chelmsford the lead and was enough to see them go into the break ahead.
Shortly after the hour Simmonds (63) struck again to extend the lead, but Michael Rose (70) gave Colchester hope with a stunning free kick a few minutes later.
An equaliser couldn't be found with Jamie Slabber (89) wrapping up a 3-1 win in the last minute leaving The U's to "focus on the league".
